Die akute lymphatische Leukämie im Kindesalter - Untersuchungen zum Zusammenhang von in vitro Resistenz und der Expression von Resistenzgenen

Drug resistance can be caused by ATP-binding-cassette (ABC)-transporters. The aim of the present study was to analyze the association between 8 ABC-transporters (MDR1, MRP1, MRP2, MRP3, MRP4, MRP5, SMRP, BCRP) and in vitro drug resistance in acute lymphoblastic leukemia. Leukemic cells of 37 children with previously untreated ALL were analyzed. The expression of the ABC-transporters was measured by TaqMan real-time PCR. In vitro drug resistance to cytosine, vincristine, thioguanine, daunorubicin, dexamethasone and prednisone was analyzed by using the methyl-thiazol-tetrazolium (MTT) assay. In vitro drug resistance was not associated with the single or simultaneous expression of the investigated ABC-transporters. MTT assays are a validated method to analyze in vitro drug resistance but they do not seem to be useful to detect resistance which is caused by drug efflux in patient samples.
