Abstract
The atypical Rho-GTPase RhoH has shown altered expression in various forms of cancer. We investigated the structural conditions, especially the switch-regions, of the interaction of RhoH with Rac2, inhibiting the stimulation of the PLCγ¬2 and the PLCβ2, as well as of the suspected Rac2-independent inhibition of the PLC-enzymes by RhoH. Therefore we created different RhoH-mutants, which were subsequently expressed in Cos-7-cells together with a PLC and eventually Rac2, measuring the phosphoinositides produced by the PLC-enzymes.
