AbstractMembers of the highly conserved and all eukaryotes ubiquitously expressed CK1 family can be involved in numerous cellular processes. Since dysregulated CK1δ has previously been linked to tumorigenesis and numerous CK1δ mutants exhibited high oncogenic potential, inhibition of CK1δ is supposed to broaden the therapeutic window for CK1δ-overexpressing or -mutated tumors. Therefore, it is important to develop new small molecule inhibitors retaining limited off-target effects, high specificity and exhibiting higher affinity toward CK1δ mutants. In the present study, the effects of established CK1δ inhibitors on CK1δ wild type and selected mutants in vitro were first characterized. Subsequently, the ability of several newly developed IWP-based inhibitors to inhibit wild type and CK1δ mutants were characterized and we furthermore analyzed their effects on growth inhibition of Hela cell lines. Our results indicate, that these compounds represent a novel promising therapeutics for personalized cancer treatment.dc.description.abstract
