AbstractThe functional ecdsysteroid receptor is a heterodimeric complex consisting of the ecdysone receptor (EcR) and ultraspiracle (USP). Ecdysteroids mediate the activity of the heterodimer to regulate transcription of genes involved in insect development. In most insects, an additional level of control is achieved by tissue specific and temporal expression of multiple EcR and USP isoforms. Functional studies characterized the ligand binding, dimerization, and transactivation properties of the USP ligand binding domain (LBD) in mutant and normal proteins from Drosophila melanogaster. The results indicated that the USP ligand binding pocket modifies the ligand binding and transactivation capabilities of the heterodimer. Further studies characterized the transcriptional capabilities of the EcR isoforms from Drosophila melanogaster and Leptinotarsa decemlineata in response to a range of steroidal and non-steroidal ecdysone agonists and juvenile hormone in a heterologous mammalian cell culture system. The species based differences observed in such studies validate the use of heterologous cell culture assays to identify species targeted insecticidal candidates.dc.description.abstract
