AbstractEscherichia coli, Corynebacterium glutamicum and Clostridium aceticum were metabolically engineered for the production of acetone. Therefore electrotransformation was performed with plasmids harboring the acetone synthesis genes from Clostridium acetobutylicum and alternative enzymes for the reaction catalyzed by CtfAB. Whereas different E. coli stains were able to produce acetone by the plasmid based acetone synthesis operons, but C. glutamicum was not able to produce acetone. Therefore a C. glutamicum gltA deletion mutant was constructed and afterwards this organism was able to produce acetone by the plasmid based acetone genes. C. aceticum was also able to produce acetone by the plasmid pIMP_adc_ctfAB_thlA with fructose or CO2 and H2 as carbon and energy source. Therefore a transformation protocol for C. aceticum was established. And the production range in C. acetobutylicum shifted towards acetone by introducing pIMP_ctfAB_thlA and pIMP_atoDA_thlA.dc.description.abstract
